The Notch signaling pathway has been shown to regulate differentiation and synaptic transmission. Therefore, this study was to investigate the function of Notch signaling pathway in the ACC during nociceptive transmission caused by neuropathy pain
.We adopt Western boltting, N- [N- (3,5-difluorophenacetyl) l-Alanyl] -S- Phenylglycine t-butyl ester (dAPT) microinjections, RNA interference targeting Notch1, hairy and enhancer of split (Hes) 1 or Hes5, electrophysiological recording and behavioral tests to verify the relationship between Notch signaling in the ACC and neuropathic pain in rats adult male Sprague Dawley .Levels of Notch intracellular domain (NICD) increased in the ACC at day 7 after chronic constriction injury (CCI) operation or spared nerve injury (SNI).
Meanwhile, the mRNA level of Notch signaling downstream effector Hes1 Hes5 mRNA increased while the rate unchanged. Injection of DAPT, a γ-Secretase (the key enzyme involved in the Notch pathway) inhibitors, ACC to significantly reversed neuropathic pain behavior. Intra-ACC injection of short hairpin RNA (shRNA) -Notch reduce the expression of NICD and lower potentiation of synaptic transmission in the ACC. In addition, the perception of pain is also addressed in mice undergoing CCI or SNI. This process is mainly mediated by Hes1 downstream effectors, but not Hes5.Based this result, activation of the Notch / Hes1 signaling pathway in the ACC participated in the development of neuropathic pain, suggesting that Notch pathway may be a new therapeutic target to treat chronic pain.

[Decrease in long chains MALAT1 non-coding RNA expression and increased expression of HSA-miR155-3p involved in the regulation of Notch signaling pathway in patients with rheumatoid arthritis]
Objective To investigate the expression of a long chain of non-coding RNA (lncRNA) metastasis associated lung adenocarcinoma transcript 1 (MALAT1) and HSA-miR155-3p in patients with rheumatoid arthritis (RA) and their relationship with the Notch signaling pathway, and explore the possibility of RA pathogenesis. Method peripheral blood of patients with RA (RA group) and healthy controls (group NC) was used to screen differentially expressed mRNA by lncRNA and high-throughput gene sequencing.
Reverse-transcription PCR is used to detect the expression of lncRNA MALAT1, HSA-miR155-3p and Notch signaling pathway receptor ligands. Results lncRNA sequencing analysis showed a total of 9158 lncRNAs are expressed differently. Gene ontology (GO) annotation of functional classification revealed that differential mRNA is expressed mainly involved in inflammatory immune response, cellular transcriptional regulation and so on.
path analysis proves that the differentially expressed mRNA was significantly associated with genes involved in rheumatoid arthritis, cellular aging, and the Notch signaling pathway. According to the prediction of cis and trans, Jagged1-2, Delta1-2 and Notch1-2 probably closely related to RA. MALAT1 the RA group was lower than in group NC, and HSA-miR155-3p expression was significantly higher than in the NC group.
