Activation of the Notch signaling pathway in the anterior cingulate cortex is involved in the pathological process of neuropathic pain

Plastic changes in the anterior cingulate cortex (ACC) is very important in hypersensitivity to pain caused by peripheral nerve injury. The Notch signaling pathway has been shown to regulate differentiation and synaptic transmission. Therefore, this study was to investigate the function of Notch signaling pathway in the ACC during nociceptive transmission caused by neuropathy pain

.We adopt Western boltting, N- [N- (3,5-difluorophenacetyl) l-Alanyl] -S- Phenylglycine t-butyl ester (dAPT) microinjections, RNA interference targeting Notch1, hairy and enhancer of split (Hes) 1 or Hes5, electrophysiological recording and behavioral tests to verify the relationship between Notch signaling in the ACC and neuropathic pain in rats adult male Sprague Dawley .Levels of Notch intracellular domain (NICD) increased in the ACC at day 7 after chronic constriction injury (CCI) operation or spared nerve injury (SNI).

Meanwhile, the mRNA level of Notch signaling downstream effector Hes1 Hes5 mRNA increased while the rate unchanged. Injection of DAPT, a γ-Secretase (the key enzyme involved in the Notch pathway) inhibitors, ACC to significantly reversed neuropathic pain behavior. Intra-ACC injection of short hairpin RNA (shRNA) -Notch reduce the expression of NICD and lower potentiation of synaptic transmission in the ACC. In addition, the perception of pain is also addressed in mice undergoing CCI or SNI. This process is mainly mediated by Hes1 downstream effectors, but not Hes5.Based this result, activation of the Notch / Hes1 signaling pathway in the ACC participated in the development of neuropathic pain, suggesting that Notch pathway may be a new therapeutic target to treat chronic pain.

[Decrease in long chains MALAT1 non-coding RNA expression and increased expression of HSA-miR155-3p involved in the regulation of Notch signaling pathway in patients with rheumatoid arthritis]

Objective To investigate the expression of a long chain of non-coding RNA (lncRNA) metastasis associated lung adenocarcinoma transcript 1 (MALAT1) and HSA-miR155-3p in patients with rheumatoid arthritis (RA) and their relationship with the Notch signaling pathway, and explore the possibility of RA pathogenesis. Method peripheral blood of patients with RA (RA group) and healthy controls (group NC) was used to screen differentially expressed mRNA by lncRNA and high-throughput gene sequencing.

Reverse-transcription PCR is used to detect the expression of lncRNA MALAT1, HSA-miR155-3p and Notch signaling pathway receptor ligands. Results lncRNA sequencing analysis showed a total of 9158 lncRNAs are expressed differently. Gene ontology (GO) annotation of functional classification revealed that differential mRNA is expressed mainly involved in inflammatory immune response, cellular transcriptional regulation and so on.

path analysis proves that the differentially expressed mRNA was significantly associated with genes involved in rheumatoid arthritis, cellular aging, and the Notch signaling pathway. According to the prediction of cis and trans, Jagged1-2, Delta1-2 and Notch1-2 probably closely related to RA. MALAT1 the RA group was lower than in group NC, and HSA-miR155-3p expression was significantly higher than in the NC group.

Description: The endoplasmic reticulum (ER) is an organelle composed of an interconnected network of tubules, vesicles and cisternae found in all eukaryotic cells. It is involved in several specialized processes such as protein translation, folding, and transport of proteins to be used in the cell membrane or secreted from the cell. Accumulation of malfolded proteins in the ER activates the unfolded protein response (UPR) and the upregulation of the ER molecular chaperones GRP78 and GRP 94. These proteins are normally bound to ER transmembrane proteins such as IRE1p and ATF6 but ER stress causes their dissociation. This allows IRE1p, a serine-threonine protein kinase, to transduce the unfolded protein signal from the ER to the nucleus. IRE1p also has an endoribonuclease activity that is required to splice X-box binding protein (XBP1) mRNA, converting it to a potent UPR transcriptional activator. ER stress also causes the cleavage of ATF6, yielding a cytosolic fragment which migrates to the nucleus and together with XBP-1, activates transcription of UPR-responsive genes. Depletion of IRE1p through the expression of a dominant negative form of IRE1p has no effect on transfected cells, but cell death via apoptosis occurs under stress conditions that cause unfolded proteins to accumulate in the ER.;;For images please see PDF data sheet

Description: The mammalian Target of Rapamycin (TOR, also known as mTOR) is an evolutionarily conserved serine/threonine kinase that regulates cell growth and cell cycle progression through its ability to integrate signals from nutrient levels and growth factors. TOR regulation is accomplished through a network of various activators and repressors. It is phosphorylated by Akt, whose activity is indirectly inhibited by the lipid phosphatase PTEN. TOR is normally associated with the regulatory proteins RAPTOR, a scaffold protein whose binding by TOR substrates is necessary for effective TOR-catalyzed phosphorylation, and GΒL, which stimulates TOR’s kinase activity towards downstream proteins. It is further regulated by the proteins Rheb, TSC1 and TSC2, which act to modulate TOR activity. Delta1 Notch ligand expression lines, Delta2 Jagged1, Jagged2 and receptor Notch1 and Notch2 in the RA group increases. Correlation analysis showed that HSA-miR155-3p is inversely proportional to MALAT1, HSA-miR155-3p is directly proportional to the Notch pathway Delta1, Jagged1, and MALAT1 is inversely proportional to Jagged2. Conclusions In patients with rheumatoid arthritis, lncRNA MALAT1 HSA-mir155-3p reduced and increased, which together regulate Notch signaling pathway changes.
