Fibulin-1, a component of the extracellular matrix (ECM), which is a prognostic, pathophysiological and diagnostic role in hepatocellular carcinoma (HCC) is still unexplored. We first found that both Fibulin-1 RNA (mRNA) or protein levels were greatly elevated in HCC tissue compared to normal tissue. Fibulin-1 correlated with overall survival is poor, and it is an independent prognostic predictor (p = 0.001). Furthermore, the Expression or Fibulin-1 inhibition is reduced or sensitized HCC cell apoptosis signal, and Fibulin-1 gag pressed HCC cells’ ability to form tumors in vivo.
Additionally, Fibulin-1 apoptosis is inhibited through the Notch while Fibulin-1 silencing no effect evident in the p-MAPK, p-c-jun and the expression of p-STAT3, and both Mcl-1 and Bcl-xL is the target of Fibulin-1. Furthermore, stromal and immune scores increased in high Fibulin-1 network and FBLN1 expression is associated with increased macrophage infiltration using xcell, TIMER and TISDIB based database tool TCGA HCC.
Importantly, RNA (cfRNA) cell-free circulating levels of Fibulin-1 in serum increased significantly in patients with HCC compared with those in healthy controls, individuals with chronic hepatitis B and patients with HBV-induced liver cirrhosis. The area under the receiver operating characteristic curve (AUC) was 0.791 for Fibulin-1, 0.640 for α-fetoprotein and 0.868 for the combination of the two tumor markers. Our findings indicate that Fibulin-1 may be a potential prognostic indicator, promising serum biomarkers and therapeutic targets in patients with HCC.

UBA6 expression and AF is a major risk factor for poor outcome after thrombolytic therapy for patients with ACI. ROC curve analysis showed that the sensitivity and specificity UBA6 well (sensitivity 100%, specificity 89%, and AUC = 0.772) which will be used to evaluate the poor prognosis of ACI. In conclusion, intensive UBA6 down-regulated MCAO-induced brain injury by inhibiting the activation of Notch signaling pathway to promote apoptosis of neurons of the hippocampus and used to predict poor prognosis of ACI.
